Residents and staff got themselves out of the Courtney Hotel in Aldersbrook Road, Manor Park, just before the London Fire Brigade arrived at 2.23am.
One woman resident was treated at the scene by paramedics from the London Ambulance Service.
Eight fire appliances, around 40 firefighters, tackled the blaze, said a London Fire Brigade spokesman.
The first and second floors of the hotel were damaged by the fire.
“37 hotel residents and staff had self-evacuated themseles from the building,” he said.

The brigade received more than 20 calls to the blaze.
Crews wearing breathing apparatus used a thermal imaging camera to check if anyone was left inside the buildingResidents in nearby houses came out to see the drama unfold and help where possible.
The blaze was put out by the crews at around 5.30 am.
The Rev Elwin Cockett, who lives nearbyl, took this picture of fire crews at the scene this morning.
He said: “We were woken by what was going on.”
rews are still at the scene.
Aldersbrook Road is still close between its junction with Empress Avenue, Manor Park and Forest Drive and buses are being diverted. Cause of te blaze is not known at this stage.
