Newham Council’s planning committee has given the green light to the proposals, which also include 'affordable' housing.
The homes will be delivered in collaboration with The Guinness Partnership.
Plans will see Plot 78 form part of the new Silo D Quarter, representing the next phase of development.
It will feature the future restoration of the Grade II listed former grain silo, Silo D, as well as a public park that will connect to the existing Millennium Mills.
Plot 78 will include two pocket parks.

The buildings will also include commercial space on the ground floor.
The regeneration of Silvertown is one of London’s largest urban redevelopment projects and will see the 60-acre docklands site, that has been mainly vacant for more than 40 years, transformed.
The project will deliver around 6,500 new homes and will also create shops, restaurants, bars and a dockside leisure hub.
Jess Wallis, project lead for Silvertown, said: "We’re making significant progress at Silvertown, and the site's immense potential becomes more evident every day.
"This latest plot of homes is a crucial part of that momentum as we work with partners to continue to prioritise the delivery of housing as early as possible in the scheme.
"The addition of the first green spaces for residents and local communities is particularly significant, especially given how long the site has remained empty."
