The Gospel Oak to Stratford and Barking lines will be shut due to planned engineering works.
There will also be no service on TfL Rail between Liverpool Street and Shenfield on Saturday and Sunday.
A rail replacement bus service to replace trains will be running for both lines throughout the weekend.
